Gabapentin drug helps people to quit cannabis..

We know that, Gabapentin (see structure; brand names FanatrexGabaroneGraliseNeurontinNupentin) is a pharmaceutical drug, specifically a GABA analogue. It was originally developed for the treatment of epilepsy, and currently is also used to relieve neuropathic pain

Now Scientists at The Scripps Research Institute have found clinical evidence that the drug gabapentin, currently on the market to treat neuropathic pain and epilepsy, helps people to quit smoking marijuana (cannabis). Unlike traditional addiction treatments, gabapentin targets stress systems in the brain that are activated by drug withdrawal.

In a 12-week trial of 50 treatment-seeking cannabis users, those who took gabapentin used less cannabis, experienced fewer withdrawal symptoms such as sleeplessness, and scored higher on tests of attention, impulse-control, and other cognitive skills, compared to patients who received a placebo. If these results are confirmed by ongoing larger trials, gabapentin could become the first FDA-approved pharmaceutical treatment for cannabis dependence.

"A lot of other drugs have been tested for their ability to decrease cannabis use and withdrawal, but this is the first to show these key effects in a controlled treatment study," said Barbara J. Mason, the Pearson Family Chair and Co-Director of the Pearson Center for Alcoholism and Addiction Research at Scripps Research. "The other nice thing about gabapentin is that it is already widely prescribed, so its safety is less likely to be an issue."

Natural products isolated from marine mollusks and sponges can reverse multidrug resistance in cancer cells


Oceans are a treasure trove of naturally produced chemical compounds with impressive and varied biological activity. One class of these compounds, the lamellarins, has some members that are cytotoxic, while others are able to reverse multidrug resistance in certain cancer cells. These lamellarins (see below structure) act by inhibiting the so-called P-glycoprotein, which enhances transport of anticancer agents out of cancer cells, thus rendering the cells multidrug resistant.

Now researchers lead by Professor Rob Capon, have set out to determine the structures of a variety of lamellarins and correlate them to the activity of the compounds. As they report in Chemistry—An Asian Journal, the activity is not determined by the core structure of the compounds but rather depends on the exact nature of pendant groups adorning the periphery of the molecule. Those compounds in which many hydroxy (OH) groups had been replaced with methoxy (OCH3) were able to reverse multidrug resistance in human colon cancer cells, while those featuring a large number of hydroxy groups showed higher cytotoxic activity.

Natural products such as the lamellarins are often valued for their cytotoxic properties for use as anticancer drugs. Capon, however, comments, "Our investigation reaffirms the view that non-cytotoxic natural products can exhibit valuable biological properties that allude to both an ecological advantage and a pharmacological potential....

Weight loss pill Qnexa wins panel vote and awaits approval

We know that, The combination of the drugs phentermine (see structure-1) and topiramate (structure -2) (trade name Qnexa) is an investigational medication for the treatment ofobesity and related conditions such as type 2 diabetes and has been found to lower blood pressure and cholesterol.  Qnexa is being developed by Vivus, a California pharmaceutical company.  Phentermine is an appetite suppressant and stimulant of the amphetamine andphenethylamine class. Topiramate is an anticonvulsant that has weight loss side effects.

Weight loss pill Qnexa wins panel vote and awaits approval: The drug was rejected in a 10-6 vote the first time it came before a Food and Drug Administration (FDA) advisory panel, in 2010, due to safety concerns. However when the medication returned for another review in February, the advisory committee gave it near-unanimous approval (20-2). Potential Method to Control Obesity: Red Wine, Fruit Compound Could Help Block Fat Cell Formation

A compound found in red wine, grapes and other fruits, and similar in structure to resveratrol, is able to block cellular processes that allow fat cells to develop, opening a door to a potential method to control obesity, according to a Purdue University study. 

While similar in structure to resveratrol -- the compound found in red wine, grapes and peanuts that is thought to combat cancer, heart disease and neurodegenerative diseases -- piceatannol might be an important weapon against obesity. Resveratrol is converted to piceatannol in humans after consumption.

"Piceatannol (see structure) actually alters the timing of gene expressions, gene functions and insulin action during adipogenesis, the process in which early stage fat cells become mature fat cells," Kim said. "In the presence of piceatannol, you can see delay or complete inhibition of adipogenesis."

Use of common pesticide, imidacloprid, linked to bee colony collapse

We know that, Imidacloprid is a systemic insecticide which acts as an insect neurotoxin and belongs to a class of chemicals called the neonicotinoids which act on the central nervous system of insects with much lower toxicity to mammals. The chemical works by interfering with the transmission of stimuli in the insect nervous system. Specifically, it causes a blockage in a type of neuronal pathway (nicotinergic) that is abundant in insects and not in warm-blooded animals (making the chemical selectively toxic to insects and not warm-blooded animals). This blockage leads to the accumulation of acetylcholine, an important insect neurotransmitter, resulting in the insect's paralysis, and eventually death. It is effective on contact and via stomach action

Immunotherapy and Chemotherapy Regimen May Prolong Survival in Advanced Cancers

Cancer patients who receive a combination of low-dose interleukin-2 and retinoic acid after conventional therapy seem to live longer than those who don't get the combination. 


Retinoic acid is derived from vitamin A. Interleukin-2, a compound that fortifies the immune system, is approved at high doses to treat "metastatic" melanoma and kidney cancer. Metastatic means that a cancer has spread.
The study showed that "these biological compounds may work at low doses. Bigger doses are not always better," said lead author Dr. Francesco Recchia, director of the oncology department at Civilian Hospital in Avezzano, Italy.
Recchia stumbled upon the possibility of using low-dose interleukin-2 (IL-2) when he switched a patient with metastatic melanoma who didn't tolerate high doses to a lower dose, and the patient had an extended response to the therapy.
This study involved 500 patients who had already responded well to chemotherapy. They had a variety of cancers, including ovarian, lung, colon, stomach, kidney, melanoma, breast and pancreatic.

Researchers with Vanderbilt-Ingram Cancer Center and the Shanghai Cancer Institute in China have discovered a possible link between a diet rich in certain vegetables and a decreased risk for breast cancer.
Corresponding author Jay Fowke, Ph.D., assistant professor of Medicine, said 3,035 women diagnosed with breast cancer were identified through the Shanghai Cancer Registry. They were closely matched with 3,037 women randomly chosen from the general population.

FDA Approves Intelence (Etravirine) for Pediatric Patients...

U.S. Food and Drug Administration (FDA) has approved Intelence (etravirine) to be administered in combination with other antiretroviral (ARV) medications for treatment of human immunodeficiency virus 1 (HIV-1) in treatment-experienced pediatric patients (6 years to <18 years old) who are experiencing virologic failure with HIV-1 strains resistant to a non-nucleoside reverse transcriptase inhibitor (NNRTI) and other ARVs.

This approval, which follows FDA priority review of the company’s supplemental New Drug Application, expands the Intelence indication and makes it the only NNRTI indicated for this use in both treatment-experienced children and adults with resistance to an NNRTI and other ARVs. The approval includes a new 25mg dose to allow for weight-based dosing in pediatric patients (6 years to <18 years old and weighing at least 16kg or 35.2 lbs). The 25mg tablet is expected to be available in the first half of May.

Antidepressant, TCP (Trabylcypromine) could help the workings of anticancer drug used in leukemia...


A new study shows that an antidepressant could be crucial in helping cancer treatment drugs reach their full potential.

The study by scientists at the Institute of Cancer Research found that tranylcypromine (TCP - cis and trans iosmers - below structures) – which can be used to treat psychotic depressive states - can make cancer cells vulnerable to the effects of a vitamin A- derivative drug called ATRA (above structure).


Retinoids are a class of chemical compounds related chemically to vitamin A. ATRA is already used successfully to treat a rare form of acute myeloid leukemia (AML), but up until now, has not been effective against other types of the disease. ATRA works by encouraging leukemia cells to mature and die naturally, but the researchers lead by Ar. Arthur Zelent say that the reason many AML patients do not respond to the treatment is because the genes that ATRA normally attacks are switched off by an enzyme called LSD1. The scientists discovered that using TCP to block this 'off switch' could reactivate these genes, making the cancer cells susceptible to ATRA.

The team has already joined forces with the University of Munster in Germany to start a Phase II clinical trial of the drug combination in AML patients. The authors also commented that both the retinoid ATRA and the antidepressant TCP are already available in the UK and off-patent, so these drugs should not be expensive for the health service.

Rigosertib drug shows promise against advanced pancreatic cancer

Researchers from University of Colorado, lead by Antonio Jimeno, have found that, a new drug, Rigosertib,  allows pancreatic cancer cells to rush through replication - and then stops them cold, killing them in in the middle of a step called M phase. Healthy cells that don't rush are unharmed.

Data from a phase I clinical trial of patients with advanced pancreatic cancer and additional solid tumors recently published in the journal Clinical Cancer Research shows the strategy has promise. While the goal of any phase I trial is to establish the dosage that best balances effectiveness against side effects, 11 of the 19 patients treated achieved stable disease, which lasted for a median of 113 days.

"Really, the drug takes one of cancer's greatest strengths and turns it into a weakness," says Wells Messersmith, MD, co-leader of the Developmental Therapeutics Program at the University of Colorado Cancer Center and the clinical trial's national principal investigator.

Instead of going with the flow of the natural cell cycle, cancer cells amplify two signals - PLK1 and PI3K - which allows them to blast through the cell cycle and divide much more quickly. In the process, they break this step of the natural cell cycle, known as the G1 regulatory mechanism, and thus depend on the kick of PLK1 and P13K to push at a frenzied pace through replication...

New drug improves glycaemic control with minimum risk of hypoglycemia in type 2 diabetics..

TAK-875, a new treatment for type 2 diabetes, improves blood sugar control and is equally effective as glimepiride, but has a significantly lower risk of creating a dangerous drop in blood sugar, called hypoglycemia, according to a new study. TAK-875 is a novel oral medication designed to enhance insulin secretion in a glucose-dependant manner, which means that it has no effect on insulin secretion when glucose levels are normal, and as such has the potential to improve the control of blood sugar levels without the risk of hypoglycemia.

In the study, Charles Burant, M.D., Ph.D., professor of internal medicine at the University of Michigan Health System, and colleagues randomly assigned 426 patients with type 2 diabetes who were not achieving adequate glucose control through diet, exercise or metformin treatment to one of five doses of TAK-875, a placebo, or glimepiride, a conventional diabetes treatment. The primary outcome was change in hemogloblin A1c from the start of the study.

At 12 weeks, all doses of TAK-875 resulted in significant drops in HbA1c compared with placebo, a similar reduction occurred in patients given glimepiride.

At a TAK-875 dose of 25 mg or higher, about twice as many patients (33 to 48 percent) reached the American Diabetics Association target of HbA1c less than 7 percent within 12 weeks, compared with placebo (19 percent) and was similar to glimepiride (40 percent).

Skin cancer drug, vemurafenib may prolong survival in advanced cases: Study

According to an international study a new treatment for advanced skin cancer almost doubles survival times. Researchers say 132 patients in the U.S. and Australia who were given the drug vemurafenib gained several extra months of life. The treatment is one of two drugs for late-stage melanoma, approved on fast-track in the US last year, which offer hope for patients with advanced melanoma. Vemurafenib is suitable for about half of patients with advanced melanoma as it targets tumors that express a certain gene mutation. Before that, there had been no new drugs for the cancer for more than a decade...

FDA panel votes in favor of earlier rejected anti-obesity drug Qnexa: Qnexa took a step closer to approval on Wednesday, when outside advisers to the U.S. Food and Drug Administration voted 20-to-2 in favor of approval. The panel also recommended the company conduct a study to scrutinize even more closely any risk of heart problems. That study may be required after the drug is approved. Should the FDA ask for further data on heart risks before any approval, it would delay Qnexa's launch further.

FDA Approves Korlym for Patients with Endogenous Cushing's Syndrome..

Korlym (mifepristone) was approved by the U.S. Food and Drug Administration to control high blood sugar levels (hyperglycemia) in adults with endogenous Cushing’s syndrome. This drug was approved for use in patients with endogenous Cushing’s syndrome who have type 2 diabetes or glucose intolerance and are not candidates for surgery or who have not responded to prior surgery. Korlym should never be used (contraindicated) by pregnant women....

FDA Approves Zioptan (tafluprost ophthalmic solution), Merck's Once-Daily, Preservative-Free Ophthalmic Medication

U.S. Food and Drug Administration (FDA) has approved Zioptan  (tafluprost ophthalmic solution) 0.0015%, the first preservative-free prostaglandin analog ophthalmic solution. Zioptan (pronounced zye-OP-tan) is approved for reducing elevated intraocular pressure (IOP) in patients with open-angle glaucoma (OAG) or ocular hypertension. Open-angle glaucoma is the most common form of glaucoma, while ocular hypertension is a condition characterized by an increase in pressure inside the eye.
